쥐의 배아 조직으로부터 슈반세포(Schwann cell)와 뉴런 세포(neuronal cell)를 각각 분리하여 각각 배양한 후에 두 세포군을 합쳐 공동배양하였다. 2주 동안 공동배양한 세포들은 myelin을 형성하였다. myelin 형성은 myelin 단백질인 myelin basic protein(MBP)의 항체를 사용하여 동초점 현미경으로 확인하였다. myelin 형성배양 세포들은 JC 바이러스를 3일 동안 감염시켰다. JC 바이러스 감염 후 myelin 형성배양 세포들은 MBP 항체를 사용하여 동초점 현미경으로 확인하였다. 그 결과 JC 바이러스 감염된 myelin 형성배양 세포들은 MBP가 거의 발견되지 않았다. 따라서 myelin 형성배양 세포들은 JC 바이러스에 의해 myelin 형성이 파괴 혹은 분해된 것으로 확인되었다. 본 연구는 과학 기술부, ICT 및 미래 계획(NRF-2017R1A2B3005753)이 자금을 지원하는 국립 연구 재단 (NRF)을 통한 기초 연구 프로그램과 연세의과대학교 교수연구 2019(CJ기업후원)의 지원을 받았다. |

(English Abstract) |
Schwann cells and neuronal cells were isolated from mouse embryonic tissues and cultured separately, and the two cell groups were combined and co-cultured. Cells co-cultured for 2 weeks formed myelin. Myelin formation was confirmed by confocal microscopy using an antibody of myelin protein, myelin basic protein (MBP). Myelin-forming cells were infected with JC virus for 3 days. Myelin-forming cells after JC virus infection were identified by confocal microscopy using MBP antibody. As a result, myelin-forming cells infected with JC virus had almost no MBP. Therefore, myelin-forming cells were found to be destroyed or degraded by the JC virus.
